Rapidly narrowing diversity to engineer proteins more efficiently

Triad, our versatile, commercial-grade computational protein design (CPD) platform, gives Protabit scientists a distinct advantage over those who apply evolutionary engineering approaches alone. Protabit’s approach starts by using Triad’s computational protein design tools to virtually explore all possible sequences for a given protein fold. Those that are predicted to be unstable or will not assume the desired fold are eliminated. Diversity is narrowed further by generating focused combinatorial libraries that are sized to match high throughput screening capacity. After screening, experimental results can be iteratively incorporated to inform new rounds of design to obtain a set of sequences enriched with the desired properties. The best variants can then be further optimized using directed evolution strategies.
